J Endod:氢氧化钙与银纳米粒子混合物抗粪肠球菌生物膜的作用

这篇研究的目的是通过激光共聚焦显微镜(CLSM)观察和评估氢氧化钙[Ca(OH)2]与0.02%银纳米粒子(AgNPs)相混合后对粪肠球菌的抗生物膜作用,并与1 mg/mL三联抗菌糊剂(TAP), Ca(OH)2和0.02% AgNPs相比较。

研究准备了90个牙本质片,消毒后孵育粪肠球菌以建立3周龄的生物膜模型。样本分别接受1 mg/mL TAP, Ca(OH)2+0.02% AgNPs混合物, Ca(OH)2或0.02% AgNPs (n=20/group)处理。每组样本再平均分为2个亚组,并孵育2和4周。未处理牙本质片(n=10)暴露于无菌生理盐水中作为阳性对照。无菌牙本质片(n=10)孵育于厌氧环境下的脑心浸出液肉汤作为阴性对照。在每个观察期尾,样本通过LIVE/DEAD BacLight<SUP>TM</SUP>染料进行染色,并通过CLSM进行分析以确定生物膜上死亡细菌的比例。采用GLM repeated measure和Tukey's tests对数据进行统计学分析(P< 0.05)。

结果显示,在2周和4周,施以1 mg/mL TAP (90.39%, 99.41%)和Ca(OH)2+AgNPs混合物(90.85%, 98.49%)样本的死亡细菌比例明显增加,分别明显高于施以Ca(OH)2 (76.14%, 91.71%)和AgNPs (62.83%, 88.07%)的样本。各组间抗生物膜的效果明显不同(P <0.05),除了1 mg/mL TAP和Ca(OH)2+AgNPs混合物(P >0.05)。所有药物在2个时间点均展现出明显的抗生物膜效果的差异。

结论:Ca(OH)2+AgNPs混合物展现出较高的抗生物膜作用,并且与1 mg/mL TAP没有明显差异。并且,根管内封药与细菌长期接触可以获得明显的抗生物膜效果。
