背景和目标:

溃疡性结肠炎(UC)与中性粒细胞浸润过多造成的附带组织损伤有关,但这种联系尚不完全清楚。由于c-MET受体酪氨酸激酶(MET)是嗜中性粒细胞趋化和响应其配体肝细胞生长因子(HGF)的细胞毒性所必需的,因此本项研究旨在鉴定HGF-MET信号传导在UC患者和小鼠肠道中性粒细胞中的功能。

方法:

研究人员收集健康对照和患有活动性(Mayo内镜下子评分2-3分)和非活动性(Mayo内镜下子评分0-1分)疾病的UC患者的血清和结肠活检,以评估血清和结肠HGF的水平。在野生型和MRP8-Cre MET-LoxP小鼠的DSS结肠炎期间评估疾病进展和免疫细胞浸润情况。

结果:

在患有活动性UC的患者以及在DSS结肠炎的炎症期的小鼠中检测到粘膜HGF表达增加。同样,活跃的UC患者血清HGF明显升高,并且与C反应蛋白和血液中性粒细胞计数呈正相关。流式细胞仪分析还表明,DSS结肠炎期间结肠MET +中性粒细胞上调。中性粒细胞中MET的基因消融降低了DSS诱导的结肠炎的严重程度。同时,T H 17细胞数量减少,这可能是由于MET缺乏性中性粒细胞减少了IL-1β的产生有关。

结论:

本项研究证实了嗜中性HGF-MET信号传导在加剧肠炎症期间的损伤中的核心作用。因此,在中性粒细胞中该途径的选择性阻断可被认为是UC中的一种新型治疗方法。
