J Endod:Axl参与了MTA诱导巨噬细胞极化的过程

人的单核细胞系THP-1培养于含有MTA溶液的培养基1, 2或3天,通过流式细胞仪检测M2巨噬细胞群体的变化。通过酶联免疫吸附测定检测M2因子的表达变化。同时也检测吞噬作用以及诱导血管发生的能力。另外,通过检测Axl, Akt, IKKα/β和IκBα的磷酸化水平评估Axl/NF-κB信号是否参与了MTA诱导细胞的作用。向施加了MTA刺激的THP-1细胞施以Axl/Akt/NF-κB信号的特异性抑制剂,检测其因子表达的变化。

流式细胞仪结果显示,MTA以时间依赖的方式增加CD206+细胞的数量。在施加MTA后,M2相关因子的表达上调。同样,MTA也增强了吞噬能力以及THP-1诱导血管发生的能力。施加MTA会通过Axl, Akt, IKKα/β, IκBα和p65的磷酸化激活Axl/Akt/NF-kB信号轴。另外,添加了特异性抑制剂会阻碍MTA诱导的白细胞介素10、转移生长因子b和血管内皮生长因子的表达。

结论:研究表明,通过M2的表型以及白细胞介素10、转移生长因子b和血管内皮生长因子的上调,MTA能够诱导巨噬细胞极化,并且Axl/Akt/NF-κB信号参与了这一过程。这些结果为MTA在临床应用中所展现出的抗感染作用提供了细胞和分子基础。
