J Periodontal Res:牙龈卟啉单胞菌感染颅骨后,破骨细胞前体的功能增强

破骨细胞的活性过高是包括牙周炎在内的炎症性骨疾病中骨吸收的主要特征。然而,除了破骨细胞与单核细胞/巨噬细胞谱系有所区别并与巨噬细胞和DC具有共同的祖细胞之外,破骨细胞前体的性质和功能还没有完全了解。

破骨细胞的活性过高是包括牙周炎在内的炎症性骨疾病中骨吸收的主要特征。然而,除了破骨细胞与单核细胞/巨噬细胞谱系有所区别并与巨噬细胞和DC具有共同的祖细胞之外,破骨细胞前体的性质和功能还没有完全了解,破骨细胞前体对体内细菌感染的反应也知之甚少。之前的研究已在证明牙周病原体牙龈卟啉单胞菌(Pg)可在体外通过核因子κB配体(RANKL)诱导破骨细胞分化的受体激活剂起作用。本实验使用Pg感染小鼠颅骨模型研究感染对破骨细胞前体调节的体内作用。

实验使用C57BL / 6野生型和髓样分化因子88基因敲除(MyD88-/-)小鼠,通过Pg感染颅骨建立感染模型。检测局部和全身性骨吸收,以及骨髓和脾脏来源的CD11b + c-fms +细胞的数量和功能。

结果表明,Pg感染可引起局部炎症浸润和破骨细胞生成,并增加骨髓和外周细胞CD11b + c-fms +破骨细胞前体的数量和破骨细胞的潜力。CD11b + c-fms + RANK +和CD11b + c-fms + RANK-是具有类似破骨细胞和促炎潜能的前体。CD11b + c-fms +细胞具有抗原特异性的T细胞免疫抑制活性,且随着Pg感染而增加。此外,MyD88参与Pg感染后破骨细胞前体的调节。

因此,Pg感染会增加前体细胞,这些前体细胞可在感染/炎症位点被转移至破骨细胞形成,同时抑制宿主的免疫反应,这有利于慢性感染和维持。了解破骨细胞前体的性质,功能和调控将有助于牙周炎在内的炎症性骨病的防治。
