精准医学刚上路 能解析的基因不到3%

每个人都有遗传密码,从上世纪90年代开始,基因组医学便以破译人的遗传密码为核心,并与疾病研究相结合逐步发展起来。近年来,医学发展非常迅猛,精准化医疗时代也逐渐到来。“这主要是得益于许多精准化医疗技术的应用。”曹雪涛院士表示,“例如测序技术、单细胞技术等对于疾病的诊断、治疗提供了很大的帮助,而且通过这些技术,使我们获得了很多以前未有和未知的数据,让我们更深入地认清生命的本质,更加准确地认识疾病发生发展的过程。”

在中国科学院院士陈润生看来,精准医学使医疗体系发生了本质变化。他表示,有了精准医学,当前的医疗从以诊断治疗为主过度到以健康保障为主。换一句话说,是从目前以患者为中心的体系转变成面向全民的医疗体系,这样的概念变化会导致大健康体系发生改变,进而会影响产业变化。

 “虽然精准医学可以推进相应产业的发展,但是精准医学才刚刚上路,大量的挑战和困难依然存在。”陈润生以基因组数据为例介绍,目前国际上对组学数据的解析能力非常有限,离基因组的完全破译还差得很远,在基因组中还存在大量的未解信息。

 “目前我们能解析的基因不到3%,这3%是基因组中遵守中心法则编码蛋白质的这部分基因信息,另外97%并不编码蛋白质,我们对这部分信息不清楚,需要大家共同深入研究。”陈润生说。

绘制单细胞图谱为疾病提供个性化治疗策略

据了解,随着精准医疗时代的到来,世界各国非常重视对单细胞研究,竞相抢占这一医学发展的前沿阵地,纷纷成立了各种各样的组织以及研究联盟。

中国医学科学院北京协和医学院副院校长郑忠伟表示,绘制人体生理、病理状态下不同组织细胞的单细胞图谱,有望为无法早期诊断及治愈的疾病提供个性化的新分子诊疗靶点及治疗策略。

 “这其中,血细胞因具有取材便利和易于准确捕获不同类型不同发育分化阶段的单个细胞的特点,适宜于作为单细胞分子图谱建立的排头兵。”中国医学科学院血液病医院(血液学研究所)常务副所院长程涛说。

为此,由中国医学科学院北京协和医学院牵头,中国医学科学院血液病医院(血液学研究所)具体负责,在中国医学科学院医学与健康科技创新工程的支持下,联合国内血液领域、免疫领域、单细胞技术以及生物信息大数据挖掘等多家单位作为成员单位,共同组建ABC 研究联盟。旨在充分发挥优势和特色,建立中国首个人类生理、病理的血细胞图谱,以期为国内外血细胞研究提供综合参考目录,从而为我国血液疾病早期诊断和治疗提供新的技术及策略,使我国在国际细胞分子图谱和血细胞研究领域实现率先理论突破和临床转化。

据介绍,ABC联盟有四大工作任务:一是科研协作。建立科研协作网络,优势互补,共同进步;二是资源共享。通过互帮互助,实现联盟内资源互转、设备共享、专家互动,建立国内血细胞组学数据网络及大数据库;三是学术交流。联盟单位联合携手,参加国际组织,联盟单位组织的国内外高层次专项培训、学术讲座,应及时相互通知,积极参加;四是人才培养。加强人才培养制度,实现联盟单位内技术骨干、学生等人员相互交流进修优先制度、联合培养制度。

 “希望联盟成立后,不仅能在基础研究上有所突破,而且能够实现临床的转化应用,推动包括血液、免疫、肿瘤、糖尿病等在内的相关领域的系统性、创新性研究。”曹雪涛院士说。
