JCLA:siRNA靶向人喉鳞癌裸鼠模型中周骨骼肌苷的抑瘤作用

近年来,喉癌的发病率呈上升趋势,但其发病机制尚不完全清楚。本研究目的是研究siRNA沉默围丝蛋白基因对人喉鳞癌裸鼠模型肿瘤的抑制作用,并探讨可能的抑制机制。

研究人员采用LSCC AMCHN8细胞系移植建立裸鼠肿瘤模型。随机将48只裸鼠分为8组,每组8只,分别用高(1.0 OD)或低(0.5 OD)剂量的围丝蛋白‐siRNA或适当的对照溶液处理。观察肿瘤生长并计算抑制率(%)。采用常规病理及电镜观察肿瘤的凋亡和增殖情况。分析了周蛋白mRNA和蛋白水平的变化。

研究显示,与未治疗组和低剂量组相比,高剂量组小鼠肿瘤生长受到明显抑制(P < 0.05)。病理检查显示治疗后小鼠肿瘤坏死及凋亡改变增加,并经过电镜证实。与对照组和低剂量组相比,高剂量组小鼠肿瘤周蛋白mRNA和蛋白表达显著降低(P < 0.05)

LSCC裸鼠模型中,周蛋白沉默与肿瘤细胞生长抑制有关。其潜在机制可能是由于受体介导的相关信号转导通路的诱导,这些信号转导通路调节肿瘤细胞生存所需的微环境。外周蛋白有望成为肿瘤治疗的新靶点。
