研究将AH Plus (Dentsply Caulk, Milford, DE), MTA Fillapex (Angelus Solucxoes Odontologicas, Londrina, Brazil)和EndoSequence BC (Brasseler, Savannah, GA)按照产品说明混匀,并制备成抽提液(final dilution: 1/10)。脂多糖(LPS) (100 ng/mL)用来诱导炎症反应。采用Water soluble tetrazolium-1 (WST-1)检验评估细胞活力。通过反转录聚合酶链式反应和实时聚合酶链式反应检测炎症介质(interleukin 6 and tumor necrosis factor alpha)和成骨标志物(ALP and OCN)基因的表达水平。通过碱性磷酸酶活性和茜素红染色评估成骨分化潜力。

结果显示,硅酸钙类封闭剂如MTA Fillapex和EndoSequence BC比AH Plus展现出更强的细胞活力。AH Plus, MTA Fillapex和EndoSequence BC减弱LPS诱导的炎症反应(P < .05)。LPS刺激减少成骨分化标志物表达水平、碱性磷酸酶活性和矿化结节的形成。然而,AH Plus和硅酸钙类封闭剂能够增加LPS刺激下的成骨分化潜力(P < .05)。

结论:硅酸钙类封闭剂展现出对MC3T3-E1细胞良好的抗炎症反应以及成骨分化潜力。
