DDS: 质子泵抑制剂会导致肠道微生物群的改变

背景
最近,有文献报道PPI会影响肠道微生物群的组成;因此,使用PPI可能会增加肠道感染的风险并导致细菌易位。在这项研究中,研究人员检测了PPI使用前后粪便微生物群组成,粪便有机酸浓度和pH值以及同一患者血液中的肠道细菌的情况。

方法
研究人员筛选了20名患有反流性食管炎的患者,他们接受了8周的PPI治疗。并对他们进行血液和有机酸浓度中的粪便微生物群组成和肠道细菌的分析,16S和23S rRNA靶向定量RT-PCR和高效液相色谱也被采用来进行相关分析。

结果
与治疗前的细菌计数相比,PPI处理后4周和8周乳杆菌种类均显着增加(分别为P= 0.011,P= 0.002)。在乳杆菌属,计数乳杆菌群,发酵乳杆菌的罗伊氏乳杆菌群和L. ruminis亚组显着治疗后4周和8周增加,与治疗前相比计数。与治疗前相比,PPI治疗后第4周和第8周链球菌种类也显着增加(P<0.01)。在PPI处理之前和之后,总有机酸浓度没有显着差异。PPI处理前后血液中细菌的检出率分别为22%和28%,无显着差异。

结论
我们的定量RT-PCR结果显示肠道生态失调是由PPI使用引起的,证实了通过宏基因组学分析获得的先前结果。
