首先,研究进行了剂量影响的评估以选择能够诱导牙髓细胞(DPCs)成牙本质分化表型的SIM生物活性浓度,并通过对其与CHSC/DPCs复合物协同作用的检测确定此剂量的可行性。将1.0 μmol/L (CHSC-SIM1.0)和0.5 μmol/L的SIM整合入CHSC中,检测细胞活力、迁移和钙沉积情况。最后,通过人造髓腔/3D培养模型体外模拟无细胞法生物材料的活性。

结果显示,0.1 μmol/L的SIM具有生物活性。它具有较强的诱导DPCs/CHSC复合物向成牙本质细胞表型分化的能力。将SIM整合入CHSC不会对细胞活性及对支架材料的粘附产生毒副作用。与对照组相比(CHSC),CHSC-SIM1.0促使支架/牙本质片上产生更高的富含钙的基质沉积。这种生物材料可以诱导DPCs从3D培养体系内向其表面迁移,并且与CHSC上的细胞相比,它可以刺激3D培养的DPCs表达更高的碱性磷酸酶、I型胶原、牙本质基质酸性磷蛋白1和牙本质涎磷蛋白。

结论:CHSC-SIM1.0能够增加DPCs的趋化性和再生潜力。
