Redox Biol:脂质组学揭示红细胞7KCh可能是心力衰竭的危险因子

心力衰竭(heart failure)是心血管疾病的一种,是由于心脏的收缩功能和(或)舒张功能发生异常而引起的。

给躯体供氧和清除体内代谢产物是心脏的主要功能之一。当心脏、血管功能发生障碍就会引起心血管疾病。脂质在心血管疾病的发病机制中扮演着重要角色。心力衰竭(heart failure)是心血管疾病的一种,是由于心脏的收缩功能和(或)舒张功能发生异常而引起的。有研究表明,红细胞脂质的变化能够指示病理生理过程发展,因此研究人员试图评估心力衰竭(HF)患者红细胞脂质谱是否具有提示疾病风险的信息。

试验表明,与正常对照组相比HF患者红细胞的脂质组成显著不同。同一HF患者红细胞中的氧化型胆固醇7-酮胆固醇(7KCh)比血浆具有更高的水平。HF患者红细胞中磷脂酰胆碱(PC),磷脂酰乙醇胺(PE)和鞘磷脂的水平降低,lysoPCs,lysoPEs和神经酰胺水平升高。从机制上看,7KCh可能促进了心肌细胞内活性氧(ROS)的形成,并通过ATF4依赖途径诱导其死亡,因此红细胞7KCh可能是HF的危险因素。

试验研究使用了液相色谱仪分析来自10个对照受试者和29个不同HF阶段的红细胞脂质体。作者又对115名对照受试者(n = 28)和患者(n = 87)进行了进一步调查也证明了上述研究发现。
