研究内容

Tybjaerg-Hansen教授为阐述脂蛋白的遗传学机制及其与动脉粥样硬化性心血管疾病的相关性作出了重大贡献。在早期英国心脏基金会的分子生物学研究小组工作中,她负责筛查基因突变患者的工作,并阐明了有家族性缺陷的载脂蛋白B-100的遗传基础。

她随后采取Mendelian随机化的方法,对带有缺血性心脏病风险的变异基因开展研究。研究最大的进展在于,确定遗传变异所致的脂蛋白(a)和甘油三酯(作为残余胆固醇标志物)水平的差异,与缺血性心脏病存在因果联系,而其他影响C反应蛋白和高密度脂蛋白胆固醇水平的变异则被证明不具有因果关系。

她的研究证明,APOC3基因变异会影响富含甘油三酯的脂蛋白形成,对缺血性心脏病风险有重要影响,这个发现对探讨高甘油三酯血症的治疗方法有重要意义。此外,她的研究小组还发现以下普通人群中存在以下几种关联,胆固醇转运蛋白基因表达与缺血性心脏病和胆石病风险之间存在联系,身高体重指数(BMI)是胆石病的因果危险因素,年龄迹象(如男性秃顶)和缺血性心脏病风险存在相关性。
