近日,刊登在国际杂志Proceedings of the National Academy of Sciences上的一篇研究论文中,来自加利福尼亚大学的研究人员在一项概念验证性实验中阐明了可以通过操控细胞交流网络的关键中心来在医学上调节相关的细胞行为,这就使得研究者可以对细胞信号网络中的部分结构进行重编程,而不是靶向作用单一的受体或细胞信号通路。

这项基础科学发现的潜在临床价值最终或可帮助开发新型技术,即药物或基因疗法来减缓或逆转疾病的恶化,比如癌症,该疾病是由沿着多种上游路径的异常细胞信号来驱动的;研究者Pradipta Ghosh博士表示,我们的研究揭示了,对细胞信号网络的中心进行可行性靶向作用或可逆转异常的细胞信号。

文章中研究者对两种蛋白片段(多肽)进行工程化操作,使其在G蛋白家族中处于关闭或开启的状态,G蛋白偶联受体通常位于细胞表面,其可以使得细胞感知并且对其周围所发生的事件产生反应,大约有30%的处方药都会通过G蛋白偶联受体来影响细胞。研究者发现,G蛋白可以在细胞内部被其它受体激活,而并不仅仅是细胞膜上,这包括一种名为GIV的蛋白,其活性参与了癌症的转移及其它疾病的发病,而且两种多肽的开启和关闭都是通过GIV蛋白受体来完成的。

在一系列的细胞培养实验中,研究者发现,机体被刮伤后,被开启功能的多肽可以加速细胞的迁移能力,而该过程和伤口愈合直接相关;相反被关闭功能的多肽则会降低癌细胞的侵略性以及减少细胞相关的肝纤维化中胶原的产生;对小鼠进行实验表明处于活性状态(被开启)的多肽可以帮助进行皮肤的快速愈合。最后研究者表示,本文研究或为后期开发新型个体化靶向疗法来改善或逆转癌症等疾病的发展提供了新的线索和希望。
