已有的研究表明,老化与免疫应答的缺陷以及全身炎症的增加有关。来自杰克逊基因医学实验室的研究人员对此进行了研究。其在PBMC,纯化的单核细胞,B细胞和T细胞中,运用系统免疫学方法,异构染色质可及性和转录组的手段,来研究这些变化背后的调控机制。

研究人员分析了来自77位年轻人和老年供体的样品,研究结果显示在PBMCs中存在新颖而强大的衰老特征。

研究人员发现在T细胞信号传导相关的启动子和增强子上存在系统染色质的关闭。同时,研究人员在静止和被抑制位点发现潜在的随机染色质开放现象。

染色质可及性和转录组的联合分析揭示了随着老化出现的免疫分子的激活/失活,并确定了IL7R基因和IL-7信号通路基因的沉默作为衰老潜在的生物标志物。

记忆CD8 + T细胞携带这一特征,其表现出老化相关的与NF-κBSTAT因子结合的损失。

因此,研究人员的结果提供了一种独特而全面的方法,来鉴定候选生物标志物,并帮助我们了解老龄化相关免疫缺陷的机理。
