使用BMP-2在大鼠腓骨缺损模型中验证骨再生的发生

在实验1、2和3中分别使用14、16和24只双侧腓骨上存在CSDs的大鼠。使用具有可吸收胶原海绵（ACS）（实验1和2）、高压灭菌自体骨（AAB）和纤维蛋白胶（FG）（实验3）以及异种骨（实验2）的BMP-2进行实验。在每个实验的随访期间进行X线和组织形态学评估。

结果显示，与对照和异种移植（猪骨）组相比，所有使用BMP-2的实验组中均观察到明显的新骨形成。虽然基于BMP载体的实验结果存在一些差异，但是在最初形成过多的骨之后，通过重塑明显减少了再生骨量。

综上所述，该研究结果表明，BMP-2由于其骨诱导性而表现出优异的骨再生能力，但是其递送系统不同效果也明显不同。ACS和FG表现出较好的骨再生能力，满足BMP载体靶向和控释的必要条件。AAB不能提供作为BMP载体的控释，但其维持空间的作用较为明显。能够为BMP/载体复合物提供足够支持的载体和支架对于大的骨缺陷是必需的，且AAB可作为有效的骨缺损修复支架来加以应用。大鼠腓骨CSD模型简单有效，可用于BMPs等药物对骨再生的初步评估。
