Wnt5aROR2SIAH2信号轴诱导与维持前列腺癌细胞在骨髓中休眠

前列腺癌居世界范围内男性恶性肿瘤第二位,高骨转移作为该肿瘤的显着特征是死亡的主要原因。临床发现,很多前列腺癌患者的骨转移发生在原发肿瘤治疗后数年,甚至十余年。对这一临床现象较为公认的解释是,肿瘤细胞在治疗前已播散至患者骨髓,并在骨髓中呈休眠状态。诱导与维持这些前列腺癌骨髓播散细胞休眠的关键机制尚有待阐明。

彭新生教授团队的研究证实,来源成骨细胞壁龛的Wnt5a蛋白通过激活非经典ROR2/SIAH2信号,抑制经典Wnt信号诱导与维持前列腺癌细胞在骨髓中休眠,是前列腺癌骨髓播散细胞休眠的关键机制之一。该研究结果回答了通过激活非经典Wnt信号抑制经典Wnt信号能否诱导与维持肿瘤细胞休眠这一重要的科学问题。该研究动物实验结果显示,应用Wnt5a能维持前列腺癌骨髓播散细胞呈休眠状态,避免其被激活形成实体肿瘤。该发现具有潜在的临床应用价值。上述研究工作得到国家自然科学基金的支持。
