在铁、炎症和药物的刺激下,经过BMP-SMAD通路,以及抑制mTOR与抑免蛋白FKBP12复合的雷帕霉素,激活了调节铁平衡的肝调素的关键调控的表达。FKBP12与BMP1型受体相互作用,避免不受控制的信号。通过药理学和基因研究,研究人员发现FKBP12具有调节铁调素的功能。在体外和小鼠肝脏细胞中,通过雷帕霉素或者他克莫司封闭FKBP12均可激活铁调素。

铁调素是一种小分子多肽,主要由肝细胞合成,作用于肝脏、小肠和网状内皮系统,传递铁代谢调节信号,调控机体铁平衡。

在野生型小鼠中,予以他克莫司紧急治疗可短暂升高铁调素。FKBP12优先作用于BMP受体ALK2。ALK2发生结合FKBP12的缺陷突变可通过BMP-SMAD信号通路上调铁调素的表达。

没有FKBP12的情况下,ALK2对非典型炎症配体激活素A发生反应。

总之,研究认为这是一种新的铁调素的调控机制,可以作为潜在治疗靶点,来增强低铁调素障碍的BMP缺陷通路信号。
