四川大学华西第二医院周圣涛课题组于2018年3月16日在开放获取期刊Genome Biology上在线发表了题为“The RNA binding protein SORBS2 suppresses metastatic colonization of ovarian cancer by stabilizing tumor-suppressive immunomodulatory transcripts”的研究论文,该研究揭示了RNA结合蛋白在卵巢肿瘤转移中的重要生物学作用,为临床肿瘤患者发展新型诊疗策略提供了理论依据。

卵巢癌是目前致死率最高的女性生殖系统肿瘤,全球每年新增病例约20万,并有11.5万卵巢癌患者死亡,晚期卵巢癌患者5年生存率不足30%。其主要原因在于卵巢癌起病隐匿,确诊时多处于晚期并伴有远处转移,且手术难以完全清除癌灶,易出现复发和耐药。然而,目前卵巢癌转移的分子机制尚不完全明确。

由于RNA结合蛋白(RNA binding proteins,RBPs)是一类能够结合不同类型RNA以及RNA的不同部位的蛋白,它们能够进而调控RNA的稳定性、表达和翻译等生物学过程。四川大学华西第二医院周圣涛课题组通过系统生物学的方法筛选了在卵巢癌恶性生物学行为中可能发生作用的一系列RNA结合蛋白,发现SORBS2分子可能在卵巢癌转移中起关键作用。

为了进一步验证该分子的生物学功能及临床相关性,研究人员进一步进行了功能验证。SORBS2的分子表达水平与卵巢癌患者的生存预后呈明显相关性,且敲除SORBS2能够显著增强卵巢癌的转移侵袭能力。其相关分子机制在于其能够结合肿瘤抑制分子WFDC1和IL-17D的mRNA,这一过程能够促进其mRNA稳定,在抑制肿瘤转移潜能的同时,还能够影响肿瘤微环境中免疫细胞的表型,进一步发挥肿瘤抑制作用。该研究结果揭示了RNA结合蛋白在卵巢癌转移中的重要作用,为卵巢癌的分子机制研究打开了一片崭新的领域,同时也为卵巢癌的临床诊断及靶向治疗提供了重要的理论依据。

该研究工作获得了国家重点研发计划,国家自然科学基金,四川省科技厅项目,华西第二医院临床科研基金等的大力支持。
