Diabetes：科学家发现脂肪氧化应激加剧代谢恶化的新机制

最近一些研究发现脂肪组织氧化应激与肥胖疾病中发生的代谢紊乱存在相关性，因为缺少能够在脂肪细胞中特异性改变氧化应激状态的小鼠模型，脂肪氧化应激与代谢紊乱在体内状态下的因果关系仍然没有得到揭示。最近来自日本的研究人员构建了脂肪特异性氧化应激清除和增强小鼠模型对氧化应激在饮食诱导肥胖相关的代谢紊乱中的作用进行了观察。相关研究结果发表在国际学术期刊Diabetes上。

在这项研究中，研究人员首先在脂肪细胞中过表达Cat和Sod1，构建了脂肪氧化应激清除模型，他们发现小鼠虽然表现出脂肪组织扩张的表型，但脂质的异常堆积减少，胰岛素敏感性得到改善。相反，在脂肪细胞中特异性清除谷胱甘肽，增强小鼠的脂肪氧化应激，小鼠会表现出脂肪组织扩张受限的情况，同时伴随着脂质的异常堆积加剧，胰岛素敏感性也发生恶化；研究结果表明在这些小鼠的白色脂肪组织中，巨噬细胞极化，组织纤维化和脂质从头合成都发生显着改变。体外研究结果发现KDM1A介导SREBF1转录活性的减弱是氧化应激抑制脂质从头合成的重要机制。
