Sci Rep:他汀类药物通过p53突变介导的细胞凋亡对肺腺癌有疗效

P53基因是一种重要的抑癌基因。p53基因突变约占所有肺癌病例的一半。越来越多的证据表明他汀类药物可通过抑制甲羟戊酸通路起到抗癌作用。

研究人员使用台湾健康保险研究数据库回顾性分析了使用他汀类药物与肺癌预后之间的相关性,主要针对早期肺癌

研究报告了他汀类药物对早期肺癌患者具有保护作用,无论化疗如何。他汀类药物治疗降低了5年死亡率(优势比0.43P<0.001)。在使用辛伐他汀治疗的突变型p53肺癌细胞中,观察到细胞凋亡水平明显更高、细胞生长被抑制和脂质筏含量被调节。此外,在p53错义突变的肺癌细胞中,辛伐他汀增强caspase依赖性凋亡通路,促进突变的p53蛋白降解,并降低运动活性。

这些数据表明,在某些肺癌患者中使用他汀类药物具有临床益处。
