国际学术期刊PLOS Genetics在线发表了中国科学院上海生命科学研究院(营养与健康研究院)计算生物学研究所汪思佳研究组名为“Genome-wide Association Studies and CRISPR/Cas9-mediated Gene Editing Identify Regulatory Variants Influencing Eyebrow Thickness in Humans”的研究论文。该论文在探索影响眉毛浓密程度的遗传因素方面取得了突破。

人类在进化中,为了更高效地进行排汗散热,褪去了身体上绝大部分的体毛,同时保留了大部分的头面部毛发。头面部毛发在人类适应进化的过程中,产生了多种不同的形态差异,研究影响这些差异的因素有助于我们进一步理解人类适应性进化的过程。然而,眉毛作为头面部毛发的一个重要组成部分,受到的研究却仍然较少。汪思佳研究组首次针对东亚汉族人群以及维族混合人群的眉毛浓度,开展了全基因组关联研究,发现了位于3号染色体3q26.33的SOX2与5号染色体5q13.2的FOXD1显着影响眉毛浓度。通过与拉美人群的数据进行荟萃分析,研究人员又发现EDAR基因同样与眉毛浓度显着相关。随后,研究人员对这三个信号区域进行效应位点精确定位(Fine-mapping),并进一步利用Crispr-Cas9靶向突变实验进行功能实验验证,结果显示rs1345417位点的G->C突变会造成SOX2表达下调,rs12651896位点区域参与调控FOXD1的表达,因此进而猜测这两个位点参与影响眉毛浓密度。以往研究发现EDAR基因参与影响毛发、牙齿、汗腺等一系列外胚层衍生物的形态,在本研究中又一次验证了EDAR基因对眉毛浓度的影响。

此外,研究人员还对眉毛浓密度进行了自然选择分析。与传统认为的结论不同,眉毛浓度并没有受到强烈的正向选择,但不排除其受到性选择的可能。

该工作揭示了影响人类眉毛浓密程度的遗传机制,也为之后拓展研究毛发密度提供了新的思路。

该项工作在汪思佳研究员、张亮研究员的共同指导下完成,主要由中科院计算生物学研究所博士研究生吴思杰、复旦大学博士张曼菲和中科院上海生科院营养代谢与食品安全重点实验室硕士杨心舟共同完成,该研究得到了科技部基础性工作专项、国家自然科学基金、中科院先导科技专项(B类)、以及马普学会等项目资助。
