类器官是一大团人工生长的细胞,可以代表人类的某些器官。类器官使得科学家们可以在人类活器官高度仿真的器官上进行研究,而不是使用真实的器官。医学科学家们还希望在将来某天,这样的研究可以带来从病人细胞长出来的类器官替代受损的器官。这个领域的研究方向之一就涉及神经细胞类器官的生长,也叫作迷你大脑。它们当然不是真实的大脑,它们不能思考、加工信息或者产生意识(至少现在还不能)。但是它们具有大脑样的结构,使得研究人员可以在临床试验之前检测药物,观察药物如何影响脑细胞。

直到最近,神经类器官的寿命都很短,主要是由于难以在营养液中保存,因为在营养液中只能保存很短时间,内部细胞就开始死亡。而真实的大脑会在毛细血管输送营养物质和氧气的情况下保持长期存活的状态。在这项新研究中,研究人员找到了方法延长类器官的存活时间,他们让这些类器官长出了毛细血管。他们发现神经类器官生存周期越长越有助于它们进一步发育,长出更大更接近真实状况的神经组织。

研究团队首先在治疗志愿者非相关疾病时提取了脑膜,然后使用方法使膜上的细胞变成干细胞,随后被编程成熟为神经细胞。同时研究团队还从相同病人身上收集了内皮细胞。这些内皮细胞被放在用于培养干细胞的液体中——干细胞在这种液体里培养了3周。然后干细胞被移出放入健康小鼠的大脑中。细胞可以在这里成熟形成人神经类器官。研究团队发现两周后毛细血管就长进了类器官内层,这就可以帮助将营养物质输送到类器官内部。
