全球首例!日本出生仅6天婴儿成功获坯胎干细胞移植

日本医生成功地将来自胚胎干细胞的肝细胞移植到一名新生儿体内,在世界上是第一次。这或许会为婴儿提供新的治疗选择。

据日本国家研究所的数据,这名新生儿患有尿素循环障碍(UCDs),肝脏无法分解有毒的氨。这种疾病一般可以通过接受肝脏移植来治疗。但是,这个当时仅有6天大的患病婴儿太小了,通常认为长到3个月至5个月大、体重约6公斤,才可以进行安全的肝脏移植。

日本国家儿童健康与发展中心(NCCHD)的医生们决定尝试“过桥疗法”,将1.9亿个来源于胚胎干细胞(ES细胞)的肝细胞注射到婴儿肝脏的血管中,直到婴儿足够大为止。

该研究所在新闻稿中说,治疗结束后“患者没有血氨浓度的增加,并能够成功完成下一阶段治疗”,即肝脏移植。

这名婴儿的性别尚未披露。目前,婴儿已从亲生父亲那里接受了肝脏移植,并在出生六个月后出院。

日本国家儿童健康与发展中心表示:“这项试验的成功证明了世界上第一个使用人类胚胎干细胞治疗肝病患者的临床试验是安全的。”

在欧洲和美国,肝细胞通常在从脑死亡的捐赠者身上移除后可以获得,但在日本的供应更加有限。这给幼儿的健康发育带来了困难,因为他们要等待长得足够大,才可以进行肝脏移植。

胚胎干细胞是从受精卵中获取的,在研究中使用它们会引发伦理问题,因为胚胎随后会被破坏。

日本国家儿童健康与发展中心是日本允许建立胚胎干细胞研究新医疗方法的两个组织之一。

根据该机构的说法,它使用的受精卵已经得到两位捐赠者的同意。
