Belghasem等人检测了结肠癌特异性VTE模型,并在下腔静脉(IVC)结扎模型中探索了一组具有血栓形成前倾向的代谢物。与对照组小鼠相比,注射了人结肠腺癌细胞的去胸腺小鼠表现出明显更高的IVC血块重量(静脉血栓形成的一种生物读数)。

对小鼠血浆进行靶向代谢组学分析发现,异种移植小鼠血中犬尿素和硫酸吲哚酚(色氨酸代谢物)水平升高,与IVC血凝块大小增加呈正相关。这些代谢物是芳基烃受体(AHR)信号的配体。因此,来自异种移植小鼠的血浆以AHR依赖性方式激活了静脉内皮细胞的AHR通路,增加了组织因子(TF)和纤溶酶原激活物抑制剂1 (PAI-1)水平。

与上述结果一致,异种移植瘤动物的IVC内皮细胞也表现出核AHR增强、TF和PAI-1表达上调,提示AHR-TF/PAI-1轴被激活。重要的是,在注射犬尿素的小鼠和异种移植瘤小鼠中,药物抑制AHR活性均可抑制TF和PAI-A在IVC内皮细胞中的表达,减小血块质量。

总而言之,本研究发现,在小鼠癌症模型中,色氨酸代谢异常;并揭示了这些代谢物和AHR-TF/PAI-1轴和VTE在癌症中的一种新的相关性。
