AUTOPHAGY:自噬缺陷诱导骨流失

在最近的一项研究中,来自西安交通大学的研究人员使用成骨细胞特异性Atg7(自噬相关7)条件性敲除(cKO)小鼠研究了ATG7在体内发育和重塑阶段的直接作用。

巨自噬/自噬是一种被高度调节的过程,涉及细胞质成分的转换,但至今为止,其在维持骨稳态中的关键作用仍然是难以捉摸的。
在最近的一项研究中,来自西安交通大学的研究人员使用成骨细胞特异性Atg7(自噬相关7)条件性敲除(cKO)小鼠研究了ATG7在体内发育和重塑阶段的直接作用。
研究人员发现,Atg7 cKO小鼠在发育和成年时都表现出骨量减少。在5月龄时,Atg7 cKO小鼠的骨小梁体积显着低于对照组。这种表型归因于成骨细胞形成和基质矿化减少,伴随着破骨细胞数量的增加,破骨细胞覆盖的骨表面的程度和TNFSF11 / RANKL(肿瘤坏死因子[配体]超家族,成员11)的分泌增加,以及TNFRSF11B / OPG(肿瘤坏死因子受体超家族,成员11b [骨保护素])的减少。
值得注意的是,成骨细胞中Atg7缺乏会引发内质网(ER)应激,而通过体内施用苯基丁酸减弱ER应激可以消除Atg7缺失介导的对成骨细胞分化,矿化能力和骨形成的影响。此外,Atg7缺乏阻碍成骨细胞矿化,并且在体外以DDIT3 / CHOP(DNA损伤诱导转录物3)和MAPK8 / JNK1(丝裂原活化蛋白激酶8)-SMAD1 / 5/8部分依赖性方式促进凋亡。重建Atg7可以改善ER应激并恢复骨骼平衡。
总之,该研究结果提供了直接的证据,即自噬在调节骨稳态中起着至关重要的作用,并提出了一种创新的治疗策略来预防骨骼疾病。
