酪蛋白激酶2血小板巨核细胞血栓形成

血小板，巨核细胞分裂出的无核细胞质，在止血和动脉血栓形成过程中发挥重要作用。酪蛋白激酶2（CK2）在巨核细胞和血小板中都可以检测到，对于CK2依赖性信号通路对巨噬细胞/血小板的病理生理机制的作用尚不明确。

血小板，巨核细胞分裂出的无核细胞质，在止血和动脉血栓形成过程中发挥重要作用。酪蛋白激酶2（CK2）在巨核细胞和血小板中都可以检测到，对于CK2依赖性信号通路对巨噬细胞/血小板的病理生理机制的作用尚不明确。

本研究对CK2调控β-亚单位对血小板生成和激活的影响进行探究。特异性敲除小鼠巨核细胞/血小板的CK2β基因（ck2β-/-）可导致巨大血小板显着减少和以及髓外巨核细胞生成增加，并伴随未成熟性血小板的比例增高。在体内，血小板的寿命无明显改变；而ck2β-/-巨核细胞微管结构异常，在骨髓中产生的碎片数量剧增，并且血小板前体的形成显着减少。ck2β-/-血小板微管聚合功能受损，导致血栓形成不能、IP3受体依赖性的细胞内Ca2+释放障碍。

推测是由于胞质Ca2+浓度增加受阻、活性依赖性α颗粒和致密颗粒分泌增加，导致ck2β-/-血小板的整合素αIIbβ3激活和聚集功能丧失。因此，在ck2β-/-小鼠，动脉形成的血栓和血栓闭塞性动脉明显减少，伴随出血时间稍微延长。除此之外，与ck2βfl/fl小鼠相比，ck2β-/-小鼠脑梗死面积显着减少、神经系统缺陷明显更少、缺血性发作后预后明显更好。

研究表明CK2β可在体内强势调控血栓形成、Ca2+依赖性血小板激活和动脉血栓形成。
