BMJ:血钙水平遗传倾向与骨密度

研究认为,在钙水平正常的个体中,血钙水平升高的遗传倾向与估计骨密度的增加无关,也没有临床相关的预防骨折的保护

近日研究人员考察了血钙水平的遗传增加是否与改善骨密度和减少骨质疏松性骨折有关。

本次研究为孟德尔随机化研究,考察了UK生物银行队列数据,采用全基因组关联Meta分析,分析了61079名个体的血清钙水平,以确定血清钙水平的遗传决定因素。英国生物银行(UK Biobank)研究的目的是评估血清钙升高的遗传倾向与超声骨密度之间的关系。研究对426824名平均钙水平在正常范围内的人进行了评估。然后进行了骨折全基因组联合Meta分析。 24个队列和英国生物数据库,包括76 549例病例和470 164名对照组,对照组的平均钙水平在正常范围内。

在逆方差加权孟德尔随机分析中,遗传衍生血钙的标准差(0.13mmol/l或0.51mg/dl)与骨密度估计值(0.003 g/cm2,95%CI:-0.059~0.066,p=0.92)或骨折风险降低(OR:1.01,95%CI:0.89~1.15,p=0.85)无关。敏感性分析未提供多同源效应的证据。
