7月14日,中国科学院上海生命科学研究院生物化学与细胞生物学研究所研究员周斌受邀在国际学术期刊《细胞研究》(Cell Research)发表了题为Cardiomyocyte proliferation: remove brakes and push accelerators 的评论文章,点评了近期同时发表于Nature上的题为The extracellular matrix protein Agrin promotes heart regeneration in mice 和Dystrophin glycoprotein complex sequesters Yap to inhibit cardiomyocyte proliferation 的两篇文章,以及发表于Cell Research上的题为Live cell screening platform identifies PPARδ as a regulator of cardiomyocyte proliferation and cardiac repair 的文章。上述三篇文章均为关于促进心脏再生的机制研究。


心肌梗死是世界上的首要致死疾病,目前,全球约有4000万患者由于心肌细胞死亡后纤维化组织替代而导致心脏衰竭。目前,心血管领域内主要通过两种方法来试图促进心脏再生。第一种方法是在心脏损伤后通过激活体内的心脏干细胞分化成为新的心肌细胞来促进心脏再生,然而,最近几个独立实验室对c-kit+心脏干细胞在体内的真实性提出质疑,引起了心血管干细胞再生医学领域的一系列争议。第二种方法侧重于在损伤后激活体内心肌细胞的增殖来修复损伤后的心脏。一般认为哺乳动物的心脏是终末分化器官,没有明显的心肌细胞增殖。但最近有研究报道,尽管心肌细胞的分裂能力非常低,每年仍然保有一定的更新速率,大约在0.3-1%的范围内。然而,这种极低的细胞更新速率显然无法产生足够的心肌细胞来修复损伤的心脏,基本没有临床治疗意义。事实上,低等脊椎动物如斑马鱼的成体心脏在心尖切除后可以再生,新生小鼠的心脏在出生后第一天切除心尖后,也具有再生能力,不过到了出生后第7天,这种再生能力就消失了。在上述这些再生模型中,新的心肌细胞主要通过原来已经存在的心肌细胞的增殖而来。基于这些研究,科学家们开始将目光更多的关注在不同阶段调控心肌细胞增殖的分子机制方面,试图找出导致不同心肌细胞增殖能力差异的关键因子,以及限制心肌细胞进入细胞周期的主要障碍。

近期,来自以色列魏兹曼科学研究所Eldad Tzahor实验室和来自德克萨斯心脏研究所的James Martin实验室同期在Nature杂志发表了有关促进心肌细胞增殖机制的研究成果,发现细胞外基质以及Yap信号通路及其二者之间的相互作用在促心肌细胞增殖方面具有重要的作用。几乎在同时,来自德国马普研究所Felix Engel实验室发表在Cell Research上一篇文章,报道PPARδ/PDK1/Akt信号通路在促进心肌细胞增殖方面具有重要的作用。

周斌的这一特邀专评点评了上述三篇文章,系统总结了上述研究成果及其相互之间的联系,并提出了新的问题和展望。
