通过测量响应于振荡流体流动的蛋白质分泌,总三磷酸腺苷(ATP)含量和细胞内钙的变化来测试完全分化的OCY454细胞的机械敏感性。观察到硬化蛋白表达和总ATP含量的增加。然而,检测到核因子κ-B配体的非常低水平的受体活化剂,并且钙响应存在很大的不一致性。然后将来自OCY454细胞的条件培养基(CM)用于培养成骨细胞和破骨细胞前体。用碱性磷酸酶(ALP)和茜素红S染色定量成骨细胞活性,而用抗酒石酸酸性磷酸酶(TRAP)染色定量破骨细胞分化。

结果显示,机械刺激的OCY454细胞释放可溶性因子,增加成骨细胞的ALP活性(p <0.05)和钙沉积(p <0.05)。当用来自流动刺激的OCY454细胞的CM处理破骨细胞前体时,大尺寸TRAP阳性破骨细胞也显著减少(p <0.05)。

该研究的结果表明,OCY454细胞响应机械负荷,释放关键因子如硬化蛋白以调节下游骨细胞,从而证明其作为体外骨细胞机械生物学研究的新细胞模型的潜力。
