J Endod:抗微生物肽LL-37对牙髓干细胞增殖和分化的影响

研究对DPSCs分别施加5和10 μg/mL LL-37后,检测了细胞活力、基因毒性、一氧化氮产量、细胞周期、牙本质涎磷蛋白(DSPP)产物以及DSPP基因的表达情况变化。

结果显示,5和10 μg/mL浓度的LL-37没有细胞毒性,并且可以普遍上调细胞活性,特别是在培养的第3天(P < .05)。然而,LL-37在任何一个浓度下都未曾明显改变一氧化氮产量。细胞周期的分析显示,10 μg/mL的LL-37在G0/G1期阻断细胞增殖(P < .05)。对照组在细胞周期的其他阶段展现出更多的细胞数量(P < .05)。同样,10 μg/mL的LL-37组DSPP基因和蛋白的表达也明显高于其他组(P < .05)。

结论:这些结果表明,LL-37在受检浓度表现出良好的生物相容性,并且它可以增加活性细胞的数量,特别是在初始期。10 μg/mL的浓度阻碍细胞周期,但是可以促进DSPP基因和蛋白的表达,表明LL-37肽可以促进成牙本质细胞分化。
