Diabetes：丁酸可以调节胰岛素抵抗肥胖小鼠肝脏线粒体功能！

丁酸及其更好的合成衍生物-FBA，可以调节线粒体功能、效率和动态性，可以认为是一种新的对抗肥胖和胰岛素抵抗的治疗策略。

脂肪肝、氧化应激和线粒体功能障碍是胰岛素抵抗和肥胖的重要病理生理特征。丁酸，由在大肠中的肠道微生物发酵产生的，作为肠道酪酸梭菌的主要代谢产物，人体内丁酸的血清半衰期仅仅6 min。 产丁酸的肠道菌群足以影响人体丁酸的供应、吸收及代谢等生理状态。丁酸的合成衍生物，N-（1-氨基甲酰-2-苯基-乙基）丁酰胺，也叫FBA，已被证明可以预防胰岛素抵抗和脂肪肝。

近日，糖尿病领域权威杂志Diabetes上发表了一篇研究文章。在这项研究中，肝脏线粒体被确定为丁酸类化合物对恢复胰岛素抵抗和在饮食诱导肥胖小鼠中脂肪积累产生有益效应的主要靶器官。特别是丁酸和FBA可以改善呼吸能力和脂肪酸氧化，激活AMPK-ACC通路，促进效率低下的代谢过程，正如增加质子泄漏。这两种治疗方法可以一致地增加底物的利用，特别是脂肪酸，导致细胞内脂质积累和氧化应激的减少。

最后，丁酸和FBA对线粒体动态融合的改变不仅导致了线粒体细胞能量代谢的改善，而且显著改善了葡萄糖稳态。

总而言之，丁酸及其更好的合成衍生物-FBA，可以调节线粒体功能、效率和动态性，可以认为是一种新的对抗肥胖和胰岛素抵抗的治疗策略。
