包含免疫受体酪氨酸抑制模体(ITIM)的受体G6b-B是血小板稳态的关键调控因子。但其具体潜在机制尚不明确。G6b-B胞质尾端的ITIM和免疫受体酪氨酸转换模体(ITSM)的酪氨酸磷酸化为包含SH2域的蛋白酪氨酸磷酸酶Shp1和Shp2提供对接位点,Shp1和Shp2也是血小板合成及其功能的重要调节因子。近日,Blood杂志上发表一篇文章,Mitchell J. Geer对G6b-B与Shp1和Shp2解偶联的生理结果进行研究。

研究人员建立表达G6B-b突变体(ITIM的212位和ITSM的238位残基酪氨酸[Y]突变成苯丙氨酸[F])的小鼠模型。纯合突变小鼠(G6b-B diY/F)患有巨血小板减少症,源于血小板合成减少;造血部位有大量巨核细胞和骨髓纤维化,与G6b敲除(G6b KO)小鼠的表现相似。由于包含免疫受体酪氨酸激活模体(ITAM)的胶原蛋白受体复合物GPVI-FcRγ链和凝血酶的表达量显著降低,G6b-B diY/F小鼠的血小板对胶原蛋白反应不灵敏,该效应可通过ADP共刺激血小板而得到部分恢复。

与此相反,G6b-B diY/F、G6b KO和巨核细胞特异性敲除Shp2小鼠的血小板对抗体介导的包含hemi-ITAM的平足蛋白受体CLEC-2的交联反应超敏,提示G6b-B可通过Shp2抑制CLEC-2介导的血小板激活。

本研究结果证明G6b-B必须与Shp1和Shp2结合,才能介导发挥其对血小板稳定的调控作用。
