用等量含/不含尼古丁(终浓度1mg / mL)的电子烟液处理HGFs,约≤48小时。 用乳酸脱氢酶测定(LDH),电子显微镜分析,I型胶原蛋白的产生,流细胞溶酶体室评估,和蛋白印迹轻链3(微管相关蛋白1A / 1B-LC3)表达来评定。

含尼古丁的电子烟液处理的HGFs中LDH水平升高,细胞质中存在大量液泡,I型胶原蛋白产生减少,II 型LC3表达增加,证实具有细胞毒性作用。无尼古丁电子烟液处理成纤维细胞的细胞质中自噬囊泡和I型前胶原分子更多。在相同的样品中,检测到溶酶体室的时间依赖性活化,LC3表达没有变化。

电子烟液(含/不含尼古丁)引起口腔成纤维细胞中分子和形态反应,引发了人们对其在口腔疾病发病机理中的作用的担忧。
