本研究将细胞生理学和磷酸化蛋白质组学方法,结合肽串联质量标签(TMT)标记、样本多路复用、同步前体选择(SPS)和三级串联质谱(MS3)来分析免疫酪氨酸活化模体(ITAM)血小板胶原受体GPVI下游的信号机制。

总体上,在GPVI介导的血小板活化的初始和进展条件下,在1300多个蛋白上检测到了3000多个显著的磷酸化事件(FDR<0.05)。通过文献指导的因果推断工具,研究人员从关键的、新兴的GPVI效应因子(如FcRg、Syk、PLCg2、PKCd、DAPP1)的磷酸化蛋白质组学数据中匹配到了300多个位点特异性信号通路关系。在相关知识背景下,GPVI/ITAM信号的调控网络也得以阐明,包括40多个Rab GTPases和相关调控蛋白系统,其中TAK1介导的Rab7 S72磷酸化与内溶酶体成熟和GPVI介导的血小板功能相关。

除了作为从组学数据集生成和检验假设的模型,本研究还提出了一种方法来识别与血栓、血管炎症和其他血小板相关疾病状态相关的止血效应物、生物标志物和治疗靶点。
