最近,有研究人员调查了患有前列腺癌(PCa)的非裔美国人(AA)全血中,DNA甲基化驱动基因的改变的临床意义。研究人员利用高通量焦磷酸测序的方法定量了PCa患者和非癌症患者全血中的8个基因(RARβ2, TIMP3, SPARC, CDH13, HIN1, LINE1, CYB5R2和DRD2)DNA甲基化水平的比例,还评估了甲基化状态和各种临床病理特性的相关性。

研究发现,在PCa和对照案例中,有6个基因在DNA甲基化水平上存在显著差异(P<0.05)。在所有分析的案例中,TIMP3位点展现出了DNA甲基化与年龄显著的相关性(p<0.05)。研究人员在CDH13甲基化和血清维生素D水平之间观察到了负相关关系(p = 0.045; r = -0.21);且在癌症案例中,TIMP3甲基化(p = 0.021; r = -0.24)和DRD2甲基化(p = 0.056; r = -0.201)表现出了与补充维生素D的负相关关系。研究人员同时还观察到了RARβ2 (p = 0.0036; r = 0.293)和SPARC(p = 0.0134; r = 0.20)位点与PSA水平在对照中具有直接的相关性,而在癌症案例中没有。另外,饮酒案例表现出了DRD2甲基化(p = 0.0349; r = -0.343)与格林森得分的负相关性。

最后,研究人员指出,他们的数据表明了在AA PCa患者的全血中,驱动基因甲基化发生的频率更高,并且与患有PCa的AA男性的各种临床病理相关。
