Blood:转铁蛋白在铁稳态和肝铁中毒中起重要的保护作用

肝转铁蛋白缺乏小鼠表现为缺铁性贫血、组织铁超载,容易发生铁中毒肝损伤。 铁死亡抑制剂或Slc39a14缺失可减轻铁超载和ccl4诱导的肝纤维化。

虽然富含血清的金属结合蛋白,转铁蛋白(由Trf基因编码),主要在肝脏中合成,但其在肝脏中的功能尚不清楚。

Yu等人建立了一种肝特异性敲除Trf的小鼠模型(Trf-LKO),这种小鼠可存活、可生育,但其红细胞生成受损、铁代谢改变。此外,用高铁饮食喂养Trf-LKO小鼠可增加它们对铁死亡诱导性肝纤维化的易感性。

重要的是,研究人员还发现用铁死亡抑制剂治疗Trf-LKO小鼠,可以有效地挽救高铁或四氯化碳(CCl4)注射引起的肝纤维化。此外,耗竭Trf-LKO小鼠的肝Slc39a14表达也可显著挽救肝铁积累,进而减轻高铁或CCl4注射引起的铁死亡诱导性肝纤维化。

此外,研究人员还发现与健康对照组相比,肝硬化患者的血清转铁蛋白和肝转铁蛋白水平明显降低,肝铁和脂质过氧化水平明显升高。

综上所述,本研究表明,肝转铁蛋白在维持肝功能中发挥保护作用,为预防铁死亡诱导性肝纤维化提供了一个可能的治疗靶点。
