成纤维细胞活化蛋白(FAP)可用于多种癌症的诊断和预后的生物标志物。FAP与prolyl寡肽酶(PREP)具有底物特异性，PREP在（神经）炎症、神经退行性变以及癌症中均有研究。目前的检测方法不能充分区分FAP和PREP，因此需要一种可靠的用于定量血浆中FAP/PREP活性比的检测方法。本研究在具有良好特性的PREP和FAP抑制剂条件下，研究人员检测了人血浆中FAP和PREP的活性。

研究人员建立了以Z-Gly-Pro-AMC为底物，在最佳条件下测定FAP和PREP活性的联合动力学分析方法。检测极限是0.009 U / L，定量极限为0.027 U / L FAP-PREP。PREP和FAP的运行内变异系数分别为3%和4%，运行间精度分别为7%和12%。通过与已建立的终点测定方法的比较，证明了该方法的准确性。溶血干扰 截止值为1.5 g / L血红蛋白。在样品制备过程中，当血小板和红细胞溶解时，PREP（而非FAP)）活性会增加。

研究表明，采用这种新方法，血浆中Z-Gly-Pro-AMC转化活性的平均67%可归因于FAP。
