Cell Metab:科学家利用细胞代谢的“脆弱性”对抗癌症

癌症干细胞是肿瘤细胞的一个亚群,与普通干细胞一样,它们能产生一系列细胞类型。调整细胞代谢能够帮助阻止癌症干细胞。

研究人员认为,癌症干细胞在癌症复发和扩散中起到关键作用,因为它们通常不受普通化疗和放疗的影响。

正如该研究的负责人Max S. Wicha博士所说:“我们使用的靶向疗法通常只在特定的一段时间内起作用,然后癌症就会变得耐药,而这种抗性正是来自于癌症干细胞,因为它们会改变自身形态以逃避靶向治疗。”

Wicha说补充道:“由于它们这种改变形态的能力,我们需要多重干细胞疗法来攻击多种形式的干细胞。”

在最新的实验中,Wicha博士分析了癌症干细胞的代谢方式,以寻找可被攻击的漏洞。研究人员在过去的研究中发现,癌症干细胞是具有灵活性的,可以在不同的状态之间进行转换:一种是保持休眠的状态,一种是快速生长的状态。状态转换能力使癌细胞得以生存和扩散。

当癌症干细胞从一种状态转变为另一种状态时,代谢尤为关键,使其成为癌症治疗的潜在靶标。当处于休眠阶段时,它们需要葡萄糖,但是当它们变得活跃并开始分裂时,它们依赖于线粒体提供的依赖于氧的能量。

为了破坏它们的代谢转换,Wicha博士及其团队利用了一种双管齐下的方法。他们首先使用阻抑制线粒体的关节炎药物,然后操纵葡萄糖阻断其代谢通路。

Wicha博士说:”我们并不是用有毒化合物来杀死细胞,而是利用细胞自身的代谢机制来消灭癌症。“

科学家们利用这种双重攻击敲除了乳腺癌小鼠模型的癌症干细胞。尽管该研究仍处于早期阶段,而且还有很多关于癌症干细胞的未解决问题,但这些研究成果还是令人鼓舞的。

由于最近有些研究发现代谢与免疫系统之间存在关联,该研究的研究人员们认为免疫疗法或许可以与代谢方法结合使用来改善癌症的治疗。
