SCIENCE:活化溶酶体使神经干细胞“返老还童”

研究人员发现,活化的NSCs中含有具有活性的蛋白酶体,而静止的NSCs中含有体积较大的溶酶体。来自年轻小鼠的静止NSCs中存在蛋白质聚集体的积聚,并且这些聚集体中的很大部分被储存在大溶酶体中。在静止的NSCs中扰动溶酶体活性能够影响蛋白质聚集体的积累,以及静止NSCs的活化能力。

在衰老的过程中,静止的NSCs显现出溶酶体方面的缺陷,出现了蛋白质聚集物的积聚增加,并且NSCs本身活化能力降低。

在老龄静止的NSCs中增强其溶酶体途径,能够帮助清除蛋白质聚集体,并改善静止NSCs活化的能力,使其恢复更年轻的状态。
