来自洛桑联邦理工学院的研究人员提供了生物信息学和实验证据,表明线粒体应激反应信号存在于涉及人类,小鼠和秀丽隐杆线虫的淀粉样蛋白蛋白毒性的疾病中。这些疾病还涉及线粒体未折叠蛋白反应和线粒体自噬过程。

研究人员使用具有淀粉样蛋白毒性的蠕虫模型,GMC101,重新测试了其线粒体特征,并证实了线粒体应激反应的诱导对维持线粒体蛋白稳态和健康至关重要。值得注意的是,通过药理学和基因靶向线粒体翻译和线粒体自噬,以增加线粒体蛋白平衡,可以增加GMC101蠕虫的适应性和寿命,并减少细胞,蠕虫和阿尔茨海默氏病转基因小鼠模型中的淀粉样蛋白聚集。
