GENE DEV:抑制自噬竟能够延长寿命

细胞自噬是机体普遍存在的分解代谢过程,其会导致细胞质成分在细胞内的降解,已有的研究表明,细胞自噬通常会对机体的健康和寿命产生积极的影响。相反,自噬的失活和细胞以及生物体的不良影响有关。根据拮抗基因多效性理论,在机体年轻的时候促进健康的一些基因在老化过程中是有害的。

细胞自噬是机体普遍存在的分解代谢过程,其会导致细胞质成分在细胞内的降解,已有的研究表明,细胞自噬通常会对机体的健康和寿命产生积极的影响。相反,自噬的失活和细胞以及生物体的不良影响有关。根据拮抗基因多效性理论,在机体年轻的时候促进健康的一些基因在老化过程中是有害的。

在此基础上,来自德国分子生物学研究所的研究人员使用RNAi筛选了影响生育后寿命的基因,并在该筛选中鉴定了30个全新的调节生育后寿命的的因子,其中就包括pha-4。研究人员对pha-4的下游因子进行了分析,他们发现了直到囊泡成核阶段的自噬早期基因的失活,包括bec-1,能够显著延长机体的寿命以及健康寿命。此外,研究人员的数据还表明,机体的健康和寿命的改善是由神经元介导的,自噬失活会导致较少的神经退行以及肌肉萎缩。

因此,研究人员认为,在年龄相关的功能障碍的背景下,细胞自噬可能从有利因素转变为有害因素。
