人血细胞分子图谱研究联盟:破译人血细胞之谜

今年9月,中国医学医科院与国内多家单位联合倡议并组建成立“人血细胞分子图谱(AtlasofBloodCells,ABC)研究联盟”,联合国内血液领域、免疫领域、单细胞技术以及生物信息大数据挖掘等多家单位作为成员单位,建立中国首个人类生理、病理的血细胞图谱。

细胞是构成生命的基因单元,系统地描绘人体生理、病理状态下不同组织细胞的单细胞图谱,对人体中所有(约37万亿个)细胞进行分类和测序,并通过这把钥匙,来加深对疾病诊断、监测、治疗的了解,这就是“人类细胞图谱计划”。

“人类细胞图谱计划”有望为现代医学无法早期诊断及治愈的疾病,提供个性化的新分子诊疗靶点及治疗策略,因此,世界各国都在竞相抢占这一未来医学发展的前沿阵地。

上月,马克?扎克伯格与其夫人普莉希拉?陈成立的陈-扎克伯格基金会(ChanZuckerbergInitiative,CZI)已经决定资助“人类细胞图谱计划”的首批38个项目。这些项目作为预研究,充当“探路人”,将对“人类细胞图谱计划”的关键问题进行探索。

在国家深入实施创新驱动发展战略和健康中国战略部署的大背景下,医学健康领域已经跨入个性化精准医疗时代。ABC的研究成果有望转化到临床应用,涵盖疾病诊断、预防等方面,比如血液恶性疾病。

汇聚力量 深耕ABC

联盟专家、中国医学科学院血液病医院(血液学研究所)常务副所长程涛在接受记者采访时介绍到,国外目前研究的是整个人体的细胞图谱,很宏大、也非常诱人,我国ABC联盟的研究方向是针对血细胞做相关突破。考虑到血细胞具有取材便利和易于准确捕获不同类型不同发育分化阶段的单个细胞的特点,适宜作为单细胞分子图谱建立的排头兵。结合我国的技术力量和科研投入,以血细胞为入口来开展研究,更能发挥基础、临床、转化为一体的科技创新研究优势。

“这是基础的、战略性的工作,也是从全局出发考虑的。”ABC联盟顾问专家、中国科学院院士陈润生这样评价ABC研究联盟。“主攻单方向的研究可以挖掘得很深,但对全局的把控能力不够强。如果有一个专门的组织、一个联盟可以把所有研究元件和因素一并考虑,那必然是为研究者们提供了很好的条件和平台。让研究人员在专注自己领域内的工作时,也能兼并其他。因此联盟是非常重要的。”

陈润生介绍,纵观我国在国际上发表的文章,已接近先进水平,但在创新性和新技术的掌握方面尚有很大进步空间。因此ABC联盟汇聚力量针对共性和关键技术难题,通过优势资源整合、知识产权共享,建立长效稳定共赢的合作机制。“ABC就是要合作。”程涛坚定地说道。

“此次联盟就是解决上述不足的一个有效手段。联盟成员单位之间要多沟通、多交流,按照联盟章程,不仅要联合国内在此领域(单细胞测序)的优势,也要涵盖国际上一些先进的实验室,进一步把实质性合作做到位。”据程涛介绍,联盟每年会举行至少一次会议,并且会按照研究主题不定期组织研讨会。“对内组织公关协作,推动国家在此领域的发展,对外发出中国声音,展示我国在此领域重要的研究成果。”

绘制ABC 助力精准医疗

近年来,医学发展非常迅猛,精准化医疗的研究炙手可热。“精准医疗的学术核心是把近年来科学上最基本最重要的科学突破转化应用到临床中,最重要的突破首先就是基因组。”陈润生表示。“基因组是个整体,而研究细胞图谱实际上是为基因组学的机体应用提供了作用方向和平台。换句话说,细胞图谱就是把组学应用到临床中需要借助的细胞水平的对象。因此要将基因组学的研究应用到临床中,需要借助的对象离不开:疾病-组织-细胞-分子。ABC的研究能更好地挖掘基因组跟疾病的关系。”

在谈到图谱的研究对疾病的治疗意义时,程涛肯定地说道,通过图谱的研究一定会发现病变细胞的变化,一定会找到有诊断价值的、有临床相关性的细胞特征,这些特征也就是机体的免疫系统在肿瘤和恶性疾病情况下发生的变化。因此图谱的研究不仅会为疾病的诊断提供精准的视角,也会打开疾病研究的崭新途径。

据了解,目前国际上针对细胞图谱的研究已有一定突破。近期,人类细胞图谱(HumanCellAtlas,HCA)联盟宣布,即将发布HCA收集的首批100万个免疫细胞的基因表达谱,为构建HCA的首个草图收集代表着几个组织和器官的至少3000万个细胞将是该联盟的一个初步的里程碑成就。这些数据会共享在资料库中,供研究人员免费使用。

ABC联盟成立之前,我国在该方向的研究就已经取得了耀眼成绩。今年6月,作为ABC联盟成员之一的北京大学生命科学学院BIOPIC中心、北京未来基因诊断高精尖创新中心张泽民研究组,已在《Cell》上发表相关细胞图谱的研究成果。研究首次在单细胞水平上描绘了肝癌微环境中的免疫图谱,发现了T细胞在肝癌患者中的特殊改变,将有助于发现针对肝癌的免疫治疗靶点。

直面竞争 理论与转化并重

据陈润生介绍:临床转化是医学研究发展的大方向,是逐渐发现、积累和突破的过程。以分子研究为基础,站在谱系的宏观角度去研究不同疾病、不同时期的变化,并将研究成果逐渐积累,建立知识库。积累到一定程度,有望实现用已经发现的知识对疾病一次性做出判断,这也就是研究突破也就是临床转化。

“ABC研究联盟刚启动不久,但发展会非常迅速。”程涛说道。应对国内外激烈的竞争,通过联盟更加紧密的合作,ABC的建立将会为国内外血细胞研究提供综合参考目录,使我国在国际细胞分子图谱和血细胞研究领域实现率先理论突破和临床转化。
