移植物抗宿主病(GVHD)是同种异体干细胞移植(SCT)后最常见的并发症。新的证据表明GVHD会导致肠道干细胞损伤。

但是,目前尚不清楚皮肤干细胞是否是皮肤GVHD的攻击靶点。Lgr5+毛囊干细胞(HFSCs)有助于卵泡发育,且在修复过程中具有再生所有类型的上皮细胞的潜能。

研究人员对Lgr5+ HFSCs在SCT后是否会受到影响进行研究,并探究可用于保护Lgr5+ HFSCs不受GVHD损伤的新的治疗方式。

研究人员发现,GVHD可减少Lgr5+ HFSCs与SCT后受损的毛囊萎缩和皮肤伤口预后的联系。局部予以皮质激素治疗(治疗多种皮肤病[包括GCHD]的标准方法),会破坏HFSCs,并不能改善皮肤稳态,即使其具有抗炎效应。相反的,Janus激酶1/2抑制剂,卢索替尼,可显著缓解SCT后的皮肤GVHD,保护Lgr5+ HFSCs,修复毛发再生和皮肤伤口预后。

本研究是首次发现GVHD会靶向损伤Lgr5+ HFSCs,局部应用卢索替尼即可保护皮肤干细胞不受GVHD损害,维持皮肤稳态。
