PLoS One:视紫红质蛋白P23H和N15S氨基酸突变的分子特性比较

英国考文垂沃里克大学医学院生物医学科学系的Mitchell J等人近日在PLoS One杂志上发表了一篇重要的研究论文。编码色素蛋白视紫红质的RHO基因突变是常染色体显性视网膜色素变性(ADRP)的最常见原因。之前对视紫红质不同结构域中ADRP突变的研究表明,导致视紫红质结构更不稳定的变化是造成患者具有更严重病症的原因。他们通过比较两个RHO突变体N15S和P23H来进一步检验这一假设,这两个突变位于N末端的结构域中。

他们首先稳定转染四环素诱导型的HEK293S细胞,在细胞中检测这两种视紫红质蛋白的体外生物化学特性,如它们的紫外-可见吸收、红外傅立叶变换、圆二色性和金属视紫红质II荧光光谱特性等。与严重受损的P23H分子功能相比,N15S在结构和稳定性方面仅是略有缺陷。他们从分子基础提出这些结构差异的主要原因:与P23相比,N15残基在紫红质折叠核心中的距离更大。

因此,如先前针对野生型视紫红质所研究发现的,添加细胞质变构调节剂二氢卟酚e6尤其是可以稳定P23H的蛋白,通常可以拯救其稳定性受氨基酸置换影响的那些ADRP视紫红质蛋白。
