精准医疗有望带来更高效肺动脉高压疗法

英国帝国理工学院10月30日发布一项研究显示,研究人员基于精准医疗技术,将一种癌症治疗药物试验性地用于治疗肺动脉高压,早期临床试验效果不错。如进一步试验顺利,未来有望为这种病症的病人开发出量身定制的有效疗法。

肺动脉高压指肺动脉压力升高超过一定界值的一种血流动力学和病理生理状态,如不加以治疗,患者通常会在数年后因心力衰竭死亡。

由帝国理工学院和加拿大艾伯塔大学研究人员领衔的团队针对肺动脉高压展开了深入研究。他们分析了20名患者的状况以及来自病人的肺组织样本。

通常,肺动脉高压病患体内的线粒体功能是紊乱的。根据报告,团队发现这类病人肺部一种名为PDK的酶含量要高于普通人,并且在线粒体中较为活跃。进一步临床试验显示,此前主要用于脑部肿瘤试验性治疗的药物二氯乙酸盐能准确以这种酶为目标并阻止它发挥作用,由此使线粒体的功能提升,让病人肺部血压降低。

但团队也指出,并不是所有病患都对这种治疗药物产生反应,这与有些病患的相关基因出现变异有关。

报告作者之一、帝国理工学院教授马丁·威尔金斯说,这是精准医疗的一个很好典范,病患本身的基因能被用来预测一种药物是否有效,通过聚焦在那些能够对药物产生反应的病患,未来能更高效、更低成本地开展临床试验。

团队下一阶段将针对那些基因上能够对二氯乙酸盐产生反应的病患开展临床试验,以便为这类病患开发出更具针对性的新疗法。
