Cell:中科院发现衰老诱发神经退行性疾病的重要分子机理

神经退行性疾病,包括阿茨海默症(AD)、脊髓侧索硬化(ALS)、额颞叶痴呆(FTD)等,都是与衰老相关的疾病。神经退行性疾病给患者以及家庭带来巨大的痛苦与负担,然而目前世界范围内还没有任何一种药物能够有效治疗神经退行性疾病。

神经退行性疾病,包括阿茨海默症(AD)、脊髓侧索硬化(ALS)、额颞叶痴呆(FTD)等,都是与衰老相关的疾病。神经退行性疾病给患者以及家庭带来巨大的痛苦与负担,然而目前世界范围内还没有任何一种药物能够有效治疗神经退行性疾病。世界卫生组织预测,到2040年,神经退行性疾病将会取代癌症,成为人类第二大致死疾病。但是目前人们并不了解衰老是如何促进神经退行性疾病的发生的。因此,衰老促进神经退行性疾病的分子机理是目前神经科学研究的重点课题之一。

神经退行性疾病与基因突变有着密切的关联,但是人们并不清楚这些基因突变是如何与衰老相互作用来诱导神经退行性疾病的发生,这为开发治疗神经退行性疾病的药物和方法带来了很大困难。

中科院上海有机化学研究所生物与化学交叉研究中心袁钧瑛课题组经过多年的研究发现,RIPK1的活化可以在神经退行性疾病小鼠模型中以及人类阿茨海默症和脊髓侧索硬化的疾病样本中检测到。同时,抑制RIPK1活性可以减轻神经退行性疾病小鼠模型中的炎症及神经细胞死亡。



8月24日,这项重要研究成果在线发表于《细胞》(Cell)杂志。袁钧瑛教授作为细胞程序性死亡领域的奠基人,长期从事细胞程序性死亡的分子机制以及神经退行性疾病的致病机理的研究。

据悉,该课题组发现了细胞死亡信号通路中的关键调节因子RIPK1在神经退行性疾病中的重要作用。RIPK1是“受体作用蛋白激酶”家族成员之一,负责细胞凋亡、细胞坏死、细胞炎症等重要生理过程的开启与调控。研究人员发现,ALS风险基因TBK1能够直接结合到细胞死亡复合物中磷酸化修饰RIPK1,进而抑制RIPK1的激活以及伴随的细胞程序性死亡。在TBK1缺失的情况下,RIPK1从被抑制的状态中得以解脱,因此在肿瘤坏死因子的刺激下更容易激活并引发细胞死亡。这一现象完美的阐释了TBK1缺失小鼠胚胎致死的原因。当RIPK1激酶活性缺失(RIPK1 D138N)的时候,TBK1缺失的小鼠便能很好的存活。RIPK1作为死亡信号调控的关键因子,其活性受到多种蛋白的调节。

研究人员通过分析发现了一个有趣的现象,即另外一个RIPK1的抑制因子TAK1在TBK1缺失的时候活性增加,增强了对RIPK1的抑制作用,从而在一定程度上弥补了TBK1缺失导致的后果。

“TBK1的基因突变是导致一部分家族性ALS/FTD的重要因素,但是大部分TBK1突变的携带者仍然在60岁之后发病,暗示了衰老在TBK1突变引发ALS/FTD的过程中起到了重要的推动作用。”袁钧瑛告诉记者,“我们重点研究了衰老是如何在TBK1部分缺失的情况下促进ALS的发生。分析了青年与老年人额叶皮层的基因表达变化,发现另一个RIPK1的抑制因子TAK1在老年人脑中表达量显著降低。”

这一现象提示了衰老会导致TAK1对于RIPK1的抑制作用逐渐降低。所以,在TBK1部分缺失的人群中,衰老导致的TAK1下降会与TBK1的缺失有叠加作用,进一步促进RIPK1的活化而导致神经退行性疾病的发生。

进一步通过小鼠实验发现,发现RIPK1的活性抑制能够很好的挽救所有的ALS/FTD疾病症状与行为学变化,证明了RIPK1的过度活化才是ALS/FTD发病的关键因素,这一发现进一步证明了RIPK1是治疗ALS/FTD的重要靶点。

中国科学技术大学教授、中国科学技术大学脑衰老及脑疾病研究中心主任申勇认为,衰老会引起包括神经系统在内的各种器官功能的退化,但并不了解在分子水平上,衰老是如何促进疾病的发生。探究机体衰老的分子机理,特别是脑衰老向神经退行性疾病演化的关键分子调控机理,是神经科学领域的前沿问题之一。他表示,这项研究首次清楚地阐释了TBK1缺失以及基因突变导致ALS/FTD发病风险增加的原因,同时也是第一次发现了衰老对于ALS/FTD发病的分子机理。
