人们对血浆中循环DNA的起源组织有很大的研究兴趣。使用DNA甲基化标记产生的数据表明，白细胞谱系的造血细胞是循环DNA库的重要贡献者。然而，目前关于红细胞谱系的细胞是否也会能将DNA释放到血浆中这一问题尚不清楚。近日，国际杂志 《CLIN CHEM》在线发表一项关于人类血浆中存在红细胞产生的DNA，并能显示贫血的类型。 研究人员使用红细胞和其他组织类型的高分辨率甲基化曲线，发现了3个基因组位点在成红细胞中呈低甲基化，而在其他细胞类型中则显示高甲基化。研究人员还开发了用于测量每个基因座的差异甲基化区域的红细胞DNA的数字PCR方法。基于铁螯合酶基因的甲基化标记，健康受试者血浆红细胞DNA的中位数为30.1％。在不同病因贫血的患者中，循环红细胞DNA的定量分析可以反映骨髓中的红细胞生成活性。对于红细胞生成活性降低的患者，如再生障碍性贫血所显示，循环红细胞DNA的百分比出现降低。对于活性增加但无效的红细胞生成的患者，如以地中海贫血症为主的例证显示，循环红细胞DNA百分比显示增加。此外，研究人员发现红细胞DNA的血浆浓度与再生障碍性贫血和缺铁性贫血的治疗反应相关。使用针对其他2个差异甲基化区域
