PNAS:H2O2 响应的脂质体纳米探针用于光声的炎症成像和肿瘤治疗

过氧化氢(H2O2)在调节多种生理过程包括细胞生长、免疫应答和衰老中发挥着重要作用。H2O2 产生的不平衡与多种疾病包括癌症、糖尿病、炎症、心血管和神经变性疾病密切相关。因此,准确和灵敏地检测 H2O2 不仅可以将其用于疾病诊断,而且有利于更好地理解疾病机制,具有显著的临床价值。

过氧化氢(H2O2)在调节多种生理过程包括细胞生长、免疫应答和衰老中发挥着重要作用。H2O2 产生的不平衡与多种疾病包括癌症、糖尿病、炎症、心血管和神经变性疾病密切相关。因此,准确和灵敏地检测 H2O2 不仅可以将其用于疾病诊断,而且有利于更好地理解疾病机制,具有显着的临床价值。然而,由于在生理环境中 H2O2 的浓度相对较低(低于 50 μM),临床上基于 H2O2 的体内成像仍然是一项具有挑战性的任务。此前已有许多课题组对检测 H2O2 的荧光探针和生物发光探针进行研究,但这些基于荧光或生物发光成像进行体内 H2O2 的检测技术往往受到光的组织穿透深度(特别是可见光)的限制,无法实现深层组织中 H2O2 的准确检测。因此,设计新型的纳米探针以实现高特异性和灵敏度的深部组织与器官中 H2O2 的精确成像显得十分重要。

光声成像是一类通过检测光吸收产生的声效应成像方式,与传统光学成像相比具有大幅度提高的成像组织穿透深度,深度可达数厘米。苏州大学刘庄教授课题组设计了一个基于脂质体的纳米探针,实现了 H2O2 高特异性和高灵敏度的体内光声成像检测。他们将辣根过氧化物酶(HRP)及其底物 2,2'- 偶氮 - 双(3- 乙基苯并噻唑啉 -6- 磺酸)(ABTS)同时装载到脂质体中,获得用于体内 H2O2 检测的 Lipo@HRP&ABTS 纳米探针。在 H2O2 的存在下,无色 ABTS 将被 HRP 催化氧化成在近红外区(NIR)具有强吸收的氧化物形式,以利于 H2O2 的光声成像检测,灵敏度可以达到低于 1 μM 的浓度。利用 H2O2 探针 Lipo@HRP&ABTS,他们可以准确地检测出由脂多糖或细菌感染诱导炎症产生的过程。将该纳米探针通过尾静脉注射到小鼠体内,利用与肿瘤细胞产生的 H2O2 之间的反应,可以实现皮下小肿瘤(小于 2 mm)以及原位脑胶质瘤的光声成像。有趣的是,局部注射 Lipo@HRP&ABTS 后,基于 H2O2 含量上的差异可以进一步通过光声成像区分转移性淋巴结与非转移性淋巴结,有望实现肿瘤手术中对淋巴结清扫的精准导航。此外,他们利用 Lipo@HRP&ABTS 对 H2O2 依赖的近红外吸光度,还实现了肿瘤特异性光热治疗。这项工作开发的 H2O2 敏感的光学纳米探针,有望在炎症的光声成像检测及肿瘤的精准诊疗中发挥潜在的应用。




该论文已在线发表于 PNAS 上,论文第一作者为刘庄教授课题组的陈倩博士,项目得到了自然科学基金委和科技部纳米技术重大专项的支持。
