最近,来自日本京都大学的研究人员证实,在具有小鼠胚胎卵巢体细胞的异种重建卵巢中的长期体外培养(~4个月)期间,hPGCLCs能够逐渐分化为卵原细胞样细胞。
hPGCLC衍生的卵原细胞显示出表观遗传重编程的标志,即全基因组DNA去甲基化,印记擦除和hPSC中异常DNA甲基化的消除,并获得减数分裂重组的直接前体状态。此外,无活性的X染色体显示出进行性去甲基化和再活化,尽管是部分的。
这些发现确立了hPSC的种系能力,并为人体外配子发生迈进了关键的一步。
