研究收集了ECR损伤处的肉芽组织(n = 5)和它们周边未受影响的无症状牙周结缔组织(n = 5)。同样,也收集了慢性根尖周炎(CP)和对照样本(n = 3)。采用定量反转录聚合酶链式反应array分析比较疾病间的88个miRNAs表达模式变化。采用Student t检验评估miRNAs表达的差异变化。通过信使RNA (miRWalk)和KEGG通路的生物学信息分析鉴定预测的靶向基因和生物/细胞功能和信号通路。

结果显示,ECR中有3个miRNAs (miR-20a-5p, miR-210-3p, and miR-99a-4p)表达显著下调,1个miRNA (miR-122-5p)表达显著上调(P < .05)。CP中有1个上调和1个下调的miRNA明显达到阈值。比较ECR和CP之间的miRNA结果发现,存在3个明显不同表达的miRNA,表明疾病的发病机制有所不同。研究检测到炎症相关的Wnt, PI3K-Akt, MAPK和骨形成相关的TGFb信号通路,并推测在ECR和CP中它们可能受到不同表达的miRNA调节。每个疾病实体都有其独特的生物学进程,如:T-和B-cell受体信号通路、破骨细胞分化和CP的胞外基质-受体相互作用以及ECR的糖胺聚糖生物合成、矿物质吸收和胰岛素信号通路。

结论:这项原理验证的体内研究表明,与CP相比,ECR有常见的以及特异的miRNA表达模式,可以用来预测矿化组织调控炎症、免疫和新陈代谢的靶向基因。
