PNAS:日本科学家在脂肪细胞中发现影响胰岛素敏感性的新基因

肥胖相关的代谢平衡受损很大程度上与脂肪组织功能失调有关,但是在肥胖疾病中脂肪组织功能失调的机制还没有得到全面揭示。最近来自日本的科学家们发现了一个在脂肪细胞中高表达的基因能够改善代谢,或可成为未来治疗肥胖及相关代谢疾病的新靶点。相关研究结果发表在国际学术期刊PNAS上。

肥胖相关的代谢平衡受损很大程度上与脂肪组织功能失调有关,但是在肥胖疾病中脂肪组织功能失调的机制还没有得到全面揭示。最近来自日本的科学家们发现了一个在脂肪细胞中高表达的基因能够改善代谢,或可成为未来治疗肥胖及相关代谢疾病的新靶点。相关研究结果发表在国际学术期刊PNAS上。

在这项研究中,研究人员希望能够找到一些新基因提供一个之前未报道过的新机制来揭示脂肪细胞代谢功能,结果他们发现Fam13a(family with sequence similarity 13, member A)是一个能够影响脂肪细胞内的胰岛素级联信号的新因子。研究结果表明Fam13a在脂肪组织中高表达,并且主要表达在成熟脂肪细胞中,相比瘦小鼠,它的表达在肥胖小鼠的脂肪组织中出现了显着的下降。

研究人员发现Fam13a能够招募蛋白磷酸酶2A(PP2A)和胰岛素受体底物1(IRS1),导致IRS1避免发生蛋白酶体途径降解。该研究进一步证明敲除Fam13a能够加重肥胖相关的代谢紊乱,而特异性激活脂肪细胞中的Fam13a可以改善代谢改变脂肪组织的胰岛素敏感性。

这项研究发现了一个通过Fam13a调节脂肪细胞的胰岛素敏感性的新机制,揭示了该基因对调节系统性代谢平衡的重要作用,表明Fam13a可能是治疗肥胖相关代谢疾病的一个潜在药物靶点。
